To track what they can't see, pilots look to the green glow of the radar screen. Now biologists monitoring gene expression, individual variation, and disease have a glowing green indicator of their own: Brown University biologists have developed a "radar" for tracking ADAR, a crucial enzyme for editing RNA in the nervous system.
The advance gives scientists a way to view when and where ADAR is active in a living animal and how much of it is operating. In experiments in fruit flies described in the journal Nature Methods, the researchers show surprising degrees of individual variation in ADAR's RNA editing activity in the learning and memory centers of the brains of individual flies.
"We designed this molecular reporter to give us a fluorescent readout from living organisms," said Robert Reenan, professor of biology and senior author of the paper, which appears Dec. 25, 2011. "When it comes to gene expression and regulation, the devil is in the details."
Biologists already know that errors in transcribing RNA from DNA can lead to improper gene expression in the nervous system and might contribute to diseases such as epilepsy, suicidal depression, and schizophrenia. More recently they've gathered evidence that ADAR is associated with disease. For instance in a study in Nature Neuroscience two months ago, Reenan and colleagues at the University of Pennsylvania described profound connections between ADAR and a model of Fragile X mental retardation in fruit flies.
Reenan said that using the new "reporter" tool to look for correlations between ADAR activity levels and behavior or disease might yield new insights into how RNA editing errors lead to such variations. But he also speculated that the mechanics of how he and his research group created the fluorescent ADAR tracking system could be adapted to someday allow therapies based on targeted RNA repair. Their reporter works by requiring ADAR to fix a purposely broken individual letter of RNA on an engineered gene.
"We're actually repairing RNA at the level of a single informational bit, or nucleotide," Reenan said. "Here we've shown we can take a mutant version of a gene and restore its function, but at the level of RNA rather than DNA."
A reporter of an editor
Reenan and third author Kyle Jay began working to create the reporter in 2006 when Jay was an undergraduate student just embarking on what would become a celebrated senior thesis at Brown. They started with a well-known tool of molecular biology: a jellyfish gene that produces a protein that glows green upon exposure to ultraviolet light. The strategy was to intentionally break the gene in a way that ADAR is uniquely suited to fix.
First they engineered the gene to include necessary "intron" code that requires a specific splicing operation to take place. Then they inserted the "stop codon" T-A-G in place of T-G-G, which causes transcription to cease, effectively preventing production of the green fluorescent protein. But before splicing occurs and when ADAR finds the stop codon U-A-G in the RNA transcript, it edits the A to an I, which restores the correct information, and translation of the whole gene proceeds as if there were no stop mutation in the DNA. So when splicing and ADAR editing occurs, neurons with the gene reporter glow green.
To see where ADAR editing and splicing were occurring, compared to just splicing alone, they also rigged up an engineered gene with the splicing requirement, but not the T-A-G codon. That would produce yellow fluorescent protein when splicing alone occurred.
Armed with their new ADAR reporter, Reenan and lead author James Jepson set out to make some biological observations in flies. One was that ADAR activity is more pronounced in certain parts of the brains of developing larvae than it is in the brains of adults. The team also found wide variation in ADAR activity in the brains of flies of similar ages from individual to individual. This was a surprise, Reenan said, because all the flies were essentially genetically identical.
A versatile new tool?
Reenan said he is confident that the ADAR reporter could be useful in more organisms than the fruit fly. The idea of creating the reporter grew out of his lab's studies of comparative genomics in a number of species. ADAR, meanwhile, is found in both invertebrates and vertebrates. In fact, in the paper the researchers describe testing the flexibility of their engineering by inserting into their engineered jellyfish gene ? destined as it was for a fruit fly ? the splicing intron of a moth.
"Thus it was, a jellyfish-moth gene chimera was crippled by mutation, and repaired by a fruit fly enzyme," Reenan said. "Rube Goldberg would be proud."
Reenan said he plans to use the ADAR reporter in flies to continue the investigation of the genes associated with Fragile X and is eager for someone who works on the disorder in mice to give it a try.
The idea of adapting this method to direct ADAR to fix mistranscribed RNA or reverse DNA damage at the RNA level in a therapeutic fashion is farther into the future. But in a sense, at least ADAR is now on the radar.

ISLAMABAD ? Pakistan's president on Tuesday warned the Supreme Court not to take action in violation of the constitution, referring to a judicial hearing into a secret memo seeking to rein in the powerful military, a scandal that threatens the Pakistani leader.
The political crisis revolves around a memo that was allegedly sent to Washington with President Asif Ali Zardari's support in May asking for help in stopping a supposed army coup following the U.S. raid that killed Osama bin Laden. Zardari has denied any connection to the memo.
Also Tuesday, gunmen killed a senior official from the government's Intelligence Bureau in the northwestern city of Peshawar, said police officer Imtiaz Khan.
No group has claimed responsibility, but the Pakistani Taliban have killed many government officials and security personnel in the last few years.
There is long-standing tension between Pakistan's civilian government and the army because the military has staged a series of coups and ruled the country for much of its 64-year history.
The government has opposed the Supreme Court's decision to open a hearing into the scandal about a week ago, saying a judicial investigation was unnecessary because parliament was already looking into the matter. The powerful army, which has denied it intended to carry out a coup and was enraged by the memo, supports the investigation.
The Supreme Court opened its hearing after receiving a petition to do so from a handful of opposition politicians ? a common practice in Pakistan.
Zardari warned Supreme Court Chief Justice Iftikhar Chaudhry to respect the constitution, an indication he may be worried the judge will team up with the president's opponents to topple the government. Zardari has clashed with both Chaudhry and the army since he was elected in 2008.
"Anyone casting a bad eye intending to break up my federation, I will not let it break," Zardari told thousands of flag-waving supporters in southern Pakistan in a speech marking the fourth anniversary of the assassination of his wife, former Prime Minister Benazir Bhutto.
Zardari did not say exactly what he meant by the Supreme Court respecting the constitution, or what he fears might be the result of the inquiry. Many analysts agree the president enjoys immunity from prosecution while in office
Zardari said earlier in the day that Pakistanis should pay tribute to his slain wife by guarding against anti-democratic conspiracies, an apparent reference to tensions over the memo scandal. He said his wife's death was also a conspiracy against Pakistani democracy.
"I therefore urge all the democratic forces and the patriotic Pakistanis to foil all conspiracies against democracy and democratic institutions," said Zardari in a statement sent to reporters.
The army-backed Supreme Court hearing sparked Prime Minister Yousuf Raza Gilani to say last week that a conspiracy was under way to topple the government. He did not specifically point to the military, but said the army must be answerable to the parliament and could not act as a "state within a state."
Gilani eventually backed away from his comments after army chief Gen. Pervez Ashfaq Kayani denied any intention to stage a coup and promised to support democracy. The prime minister on Monday denied reports he would replace Kayani or the army's intelligence chief, Lt. Gen. Shuja Pasha, to neutralize the threat to his government.
Former Pakistani ambassador to the U.S., Husain Haqqani, allegedly crafted the memo sent to Washington, which promised to replace Pakistan's national security hierarchy with people favorable to the U.S. in exchange for help in reining in the military. Haqqani has denied any connection to the memo but resigned in the wake of the scandal.
The bin Laden operation angered Pakistani officials because they weren't told about it beforehand and humiliated the army because it was not able to stop the nighttime raid near Pakistan's equivalent of West Point.
The political crisis comes at a time when Pakistan is struggling with a violent Taliban insurgency, a stuttering economy and troubled relations with its most important ally, the United States.

After more than 20 years in theater management, Frank Wood made the leap from show business to business academia right here in Key West.
As executive director of the Tennessee Williams Theatre, located at Florida Keys Community College's Key West campus, Wood has been surrounded by collegiate life for the past seven years.
Eventually, his regular interactions with college students and faculty sparked a new career endeavor -- on just the other side of the campus.
"I just had this desire: I want to teach," said Wood, who joined the FKCC faculty in 2010 and now oversees the business program, which includes an Associate in Applied Science degree in business administration and a certificate in entrepreneurship.
It's been a marriage made in heaven, according to Wood, who still runs the theater from a convenient distance.
"Both my jobs are things that I really like doing," he said. "I get to feed both sides of my brain."
Wood holds a bachelor's degree and an MBA in marketing from Mercer University and is in the final stages of a doctoral program in marketing at Nova Southeastern University.
Wood's educational background, combined with his creativity and business experience, including the development and management of Keystix.com, enable him to teach business in a way that is stimulating and relevant.
"I want my students to see that there are many different ways to get to the top other than just getting an Ivy League MBA and immediately running a company," said Wood. "That's not always going to be the case for our students, and I want them to see there are different beginnings that lead to great successes."
Wood is looking forward to teaching entrepreneurship for the first time next semester.
The course is the capstone of the college's new 12-credit entrepreneurship certificate, which Wood helped to develop with valuable insight and expertise from local business leaders who comprise the program's advisory council.
"The students learn these wonderful skills -- like how to dive or maintain a boat -- but may not have the business acumen to turn it into a business," said Wood. "We teach them how to take their passion and turn it into something that can be a livelihood."
Wood and his partner, Terry Lienhardt, live on Sugarloaf Key, where they have three Rhodesian ridgeback dogs.
Wood is also an accomplished pianist and has performed regularly in some of Key West's finest restaurants, bars, and at the Metropolitan Community Church.

It's looking pretty clear that the new Democratic majority on the Wake County school board will not be retaining the legal services of Republican attorney Kieran Shanahan.
Shanahan's legal contract came up last week during the school board's discussion of a proposal to increase Tharrington Smith's hourly legal rate by $5. During the work session discussion, it was noted that Shanahan's law firm is still under contract through June to receive up to $25,000.
The new unanimously approved contract will pay Tharrington Smith's partners $190 per hour, $175 an hour for associates and $95 per hour for paralegals. It's roughly a 2.7 percent increase for partners and 2.9 percent for associates.
Chief Business Officer David Neter said that the rates Tharrington has been charging Wake are two to three times less than what law firms typically charge.
Board members from both political parties praised Tharrington Smith's work, saying they had no problems with the rate increase. A prior legal audit done by Thomas Farr had praised Tharrington Smith's work but suggested hiring an in-house counsel to replace the law firm.
Neter said that Charlotte-Mecklenburg, which has an in-house school attorney, has sharply higher legal bills than Wake.
School board member Jim Martin asked how much it would cost for this mid-year increase for Tharrington Smith. Neter said that, based on usage remaining the same, about $13,000 more this fiscal year.
Then Martin pointed to the contract adopted by the prior board with the Shanahan Law Group.
Shanahan was hired for up to $25,000 to perform various legal services, such as working on the redistricting plan and the Open Meetings lawsuit. The firm has only been paid $370 so far this fiscal year as most of the work was done before July.
Martin suggested coming up with the money for Tharrington by taking the roughly $13,000 encumbered to cover potential costs for the rest of the fiscal year for Shanahan.
School board member Chris Malone argued they should leave the money there just in case they need the flexibility of using Shanahan over the next six months.
During the discussion, board member Susan Evans asked if Farr was still under contract. She was told the answer was no.
The board wound up approving the Tharrington Smith rate increase without taking the funding from Shanahan. But the implication is clear that the contract won't be renewed during the annual meeting in June.
Shanahan and attorneys from his firm gave money to the Republican school board candidates during the campaign.

A series of strong earthquakes struck the New Zealand city of Christchurch on Friday, rattling buildings, sending goods tumbling from shelves and prompting terrified holiday shoppers to flee into the streets. There was no tsunami alert issued and the city appeared to have been spared major damage.
One person was injured at a city mall and was taken to a hospital, and four people had to be rescued after being trapped by a rock fall, Christchurch police said in a statement. But there were no immediate reports of serious injuries or widespread damage in the city, which is still recovering from a devastating February earthquake that killed 182 people and destroyed much of the downtown area.
The first 5.8-magnitude quake struck Friday afternoon, 16 miles (26 kilometers) north of Christchurch and 2.5 miles (4 kilometers) deep, the U.S. Geological Survey said. Minutes later, a 5.3-magnitude aftershock hit. About an hour after that, the city was shaken by another 5.8-magnitude temblor, the U.S.G.S. said, though New Zealand's geological agency GNS Science recorded that aftershock as a magnitude-6.0. Both aftershocks were less than 3 miles (5 kilometers) deep.
The Pacific Tsunami Warning Center did not issue an alert.
The city's airport was evacuated after the first quake and all city malls shut down as a precaution.
About 60 people were treated for minor injuries, including fractures, injuries sustained in falls and people with "emotional difficulties," Christchurch St. John Ambulance operations manager Tony Dowell told The Associated Press.
"We have had no significant injuries reported as a result of the earthquakes today," he said.
Warwick Isaacs, demolitions manager for the Canterbury Earthquake Recovery Authority, said most buildings had been evacuated "as an emergency measure." The area has recorded more than 7,000 earthquakes since a magnitude-7.0 quake rocked the city on Sept. 4, 2010. That quake did not cause any deaths.
Rock falls had occurred in one area and there was liquefaction ? when an earthquake forces underground water up through loose soil ? in several places, Isaacs told New Zealand's National Radio.
"There has been quite a lot of stuff falling out of cupboards, off shelves in shops and that sort of thing, again," he said.
Isaacs said his immediate concern was for demolition workers involved in tearing down buildings wrecked in previous quakes.
"It ... started slow then really got going. It was a big swaying one but not as jolting or as violent as in February," Christchurch resident Rita Langley said. "Everyone seems fairly chilled, though the traffic buildup sounds like a beehive that has just been kicked as everyone leaves (the) town (center)."
The shaking was severe in the nearby port town of Lyttelton, the epicenter of the Feb. 22 quake.
"We stayed inside until the shaking stopped. Then most people went out into the street outside," resident Andrew Turner said. "People are emotionally shocked by what happened this afternoon."
Around 26,000 homes were without power in Christchurch, after the shaking tripped switches that cut supplies, Orion energy company CEO Rob Jamieson said.
"We don't seem to have damage to our equipment," he said. "We hope to have power back on to those customers by nightfall."
Hundreds of miles of sewer and fresh water lines have been repaired in the city since the February quake.
One partly demolished building and a vacant house collapsed after Friday's quakes, police said.
Central City Business Association manager Paul Lonsdale said the quakes came at the worst possible time for retailers, with people rushing to finish their Christmas shopping.
Despite the sizable quakes, there was no visible damage in the central business district, where 28 stores have reopened in shipping containers after their buildings were wrecked by the February quake, he said.
"Hopefully tomorrow we'll be feeling a little bit better again and restoring our faith in the will to live and to stay in Christchurch," the city's deputy mayor, Ngaire Button, told National Radio.

WASHINGTON ? House Republicans, on the eve of Tuesday?s vote denying tax relief to 160 million Americans, huddled in a conference room in the Capitol basement for more than two?hours.
Were they puzzling over how to explain to constituents why they were effectively ordering a tax increase on the middle class after fighting for much larger tax breaks for the wealthy? Were they justifying the killing of a bipartisan compromise that had the support of all but seven Senate Republicans and the tacit approval of House Speaker John?Boehner?
Nope. Turns out they were talking Monday night about their favorite scenes from ?Braveheart.? About 10 House Republicans went to the microphones to share their memories of the Mel Gibson film, Republican sources told my Washington Post colleagues Paul Kane and Rosalind?Helderman.
One member spoke about the apocryphal scene in which the 13th-century Scottish rebel William Wallace ordered his troops to moon the?English.
Another member recounted the scene in which Wallace commanded the rebels to hold their positions before raising their spears against the charging English?cavalry.
This inspired the assembled lawmakers to chant: ?Hold! Hold! Hold!?Hold!?
Finally, toward the end of the meeting, Rep. Rob Bishop of Utah bravely rose to tell his colleagues that he hated the film. He introduced a motion that all references to ?Braveheart? be banned. His colleagues laughed and heckled. The motion was not?adopted.
But Bishop was right: ?Braveheart? is a conspicuously poor choice for the House?GOP.
For one thing, the Republicans are, if anything, in a reverse- ?Braveheart? position: In this fight, they are the nobles putting down the overtaxed peasants. For another, the Scots they are emulating were defeated and slaughtered, and Wallace was captured (possibly betrayed by his own side), then drawn and?quartered.
That the House Republicans would embrace a doomed cause and its martyred leader gets at their main problem in the majority: They?d rather make a point than govern the country. And in this case, it?s not entirely clear what point they?re trying to?make.
Is it making sure the tax cut is paid for? For the last decade, Republicans approved billions of dollars in tax cuts, mostly for the rich, without paying for?them.
Is it because they want the tax-cut extension to be for a year rather than just two months, as the Senate approved? Then why did so many Republicans originally criticize any tax-cut?extension?
In killing the Senate compromise, which passed 89 to 10, with 39 Republican votes, the House GOP resorted to a variant of the ?deem and pass? resolution they derided when Democrats proposed it during the health care fight. Reneging on their pledge to hold a vote on the Senate compromise, Braveheart Republican leaders ordered up a resolution that rejected the Senate measure without a direct?vote.
Caucus chairman Jeb Hensarling of Texas, demanding a conference between the House and Senate to resolve differences, instructed his colleagues to ?go back and watch the ?Schoolhouse Rock? video? to see how ?differences are settled between the House and Senate.? But this ignored the fact that Senate Democrats had already compromised with Senate Republicans; Hensarling was asking them to compromise on their?compromise.
House Democrats didn?t exactly distinguish themselves, either. Rep. Jim McGovern of Massachusetts said Republicans had imposed ?martial law.? Rep. Jim McDermott of Washington brought a Christmas stocking and lump of coal to the floor. Minority Leader Nancy Pelosi of California recalled a Woody Allen joke (?the food at this place is really terrible ? and such small portions?) that she attributed to Yogi?Berra.
But that didn?t hold a torch to the Republicans? ?Braveheart? performance. It wasn?t the first congressional invocation of the film (Dick Gephardt once showed up to a meeting in William Wallace attire when he was House Democratic leader), but until now it hasn?t been embraced quite so?earnestly.
?Look, this is a ?Braveheart? moment,? Rep. Phil Gingrey, R-Ga., said on Fox News on Monday, describing the House Republicans? instructions to Boehner. ?You, Mr. Speaker, are our William Wallace. Let?s rush to the?fight.?
Apparently plenty of others felt the same way. Staffers emerged from the GOP caucus meeting at 6:45 p.m. Monday to say the meeting would break up in five minutes. But the Republicans? impromptu movie night didn?t end until 8:17 p.m., when Boehner, face as orange as Mel Gibson?s was blue, marched forth with his Bravehearts in a cloud of cigarette smoke toward their inevitable?tragedy.

A fundamental problem that has puzzled generations of scientists has finally been solved after more than 70 years.
An international team of scientists has discovered a subtle electronic effect in magnetite ? the most magnetic of all naturally occurring minerals ? causes a dramatic change to how this material conducts electricity at very low temperatures.
The discovery gives new insight into the mineral in which mankind discovered magnetism, and it may enable magnetite and similar materials to be exploited in new ways.
The research, published in Nature, was led by the University of Edinburgh in collaboration with the European Synchrotron Radiation Facility (ESRF) in Grenoble, France, where the experiments were conducted.
The magnetic properties of magnetite have been known for more than 2000 years and gave rise to the original concepts of magnets and magnetism. The mineral has also formed the basis for decades of research into magnetic recording and information storage materials.
In 1939, Dutch scientist Evert Verwey discovered that the electrical conductivity of magnetite decreases abruptly and dramatically at low temperatures. At about 125 Kelvin, or minus 150 degrees Celsius, the metallic mineral turns into an insulator. Despite many efforts, until now the reason for this transition has been debated and remained controversial.
When the team of scientists fired an intense X-ray beam at a tiny crystal of magnetite at very low temperatures, they were able to understand the subtle rearrangement of the mineral's chemical structure. Electrons are being trapped within groups of three iron atoms where they can no longer transport an electrical current.
Dr Jon Wright of the ESRF said: "Our main challenge was to obtain a perfect crystal, which meant using one that was tiny, just half the diameter of a human hair. Then we needed to observe subtle changes in this microscopic sample as we lowered the temperature. In Europe, this is only possible at the ESRF, thanks to the extremely high energy of its synchrotron X-rays."
Professor Paul Attfield, of the University of Edinburgh, said: "We have solved a fundamental problem in understanding the original magnetic material, upon which everything we know about magnetism is built. This vital insight into how magnetite is constructed and how it behaves will help in the development of future electronic and magnetic technologies."

RALEIGH, N.C. (AP) ? Former presidential candidate John Edwards says he has been diagnosed with a medical condition that would make it difficult for him to attend his approaching criminal trial over campaign finances and is asking for it to be delayed.
In a motion filed Thursday, Edwards' lawyers asked a federal judge to delay the start of the Jan. 30 trial for at least two months. They did not disclose his illness and filed sealed records with the court.
"The Defendant has a medical issue ... that will prevent a trial of this matter during the January 2012 Criminal Term," the motion says. "The failure to grant a continuance would be likely to result in a miscarriage of justice."
Members of the defense team could not immediately be reached for comment.
Federal prosecutors did not take a position on the medical issue.
However, prosecutors filed a separate motion opposing another Edwards request for more time to prepare. Prosecutors said they are ready to try Edwards on six felony and misdemeanor counts related to nearly $1 million from wealthy donors used to help hide his pregnant mistress during his 2008 run for the While House.
Edwards, a Democrat, denies knowingly violating the law and has pleaded not guilty.
The start date of the trial has already been put back once after Edwards said he needed more time to prepare his defense and attend his daughter's wedding.
Edwards appeared healthy last week at a pretrial hearing in the case.

In the context of Operation SnowGlobe, that statement is really the key. What we plan to do is build demand for products that people don't want -- yet. Products that will make a huge difference in preserving the frosty winters of our North Pole.
But, with the upcoming holidays in mind, I tend to stick to fulfilling the wishes I've been sent, rather than trying any last minute surprises. Ho ho ho!
